SCIENTIFIC RESEARCH EXCERPTS:

Anti-H5N1/Bird Flu Virus Flavonoids

Abstract: Methanolic extracts of this rare plant produced phytonutrients tested for their in vitro antiviral activity against highly pathogenic avian influenza strain H5N1 using plaque inhibition assay in Madin–Darby canine kidney. The results indicated that the extract possessed potent antiviral activity (100% inhibition at the concentration of 1 μg/ml). From the results, phytonutrients 1, 2, and 3 were found to inhibit the virus with a concentration of 1 ng/ml each by percentages of 68.13%, 79.66% and 73.22%, respectively, which showed a potency comparable with that of the clinically used antiviral drug Zanamivir which had a complete inhibition but at a higher concentration of 10 ng/ml compared with 1 ng of phytonutrients 1, 2, and 3.

Anti-H5N1/Bird Flu Virus Cellular Enzyme

Abstract: Highly pathogenic H5N1 infections are often accompanied by excessive pro-inflammatory response, high viral titer, and apoptosis; as such, the efficient control of these infections poses a great challenge. Forced cellular enzyme expression significantly inhibited the H5N1-induced increase in reactive oxygen species, decreased pro-inflammatory response, prevented p65 and p38 phosphorylation, and impeded viral ribonucleoprotein nuclear export and viral replication. The designated cellular enzyme overexpression also rescued H5N1-induced cellular apoptosis and alleviated H5N1-caused mitochondrial dysfunction.

Anti-SARS-CoV-2/COVID Inhibitors N-Acetylcysteine (NAC) and Glutathione (GSH)

Abstract: Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spike protein binds to the cellular receptor angiotensin converting enzyme-2 (ACE2) via its receptor-binding domain (RBD). Consistently, in vitro binding of RBD and ACE2, spike-mediated cell-cell fusion, and pseudotyped viral infection of VeroE6/TMPRSS2 cells were inhibited by the thiol-reactive compounds N-acetylcysteine (NAC) and a reduced form of glutathione (GSH). Furthermore, we demonstrated that the activity of variant spikes from the SARS-CoV-2 alpha and delta strains were also suppressed by NAC and GSH.
